Bacteriophage Display
1985Discovery that a foreign peptide fused to a filamentous phage's minor coat protein (gene III) is displayed on the phage surface while remaining infectious -- turning a phage's natural infection/coat-assembly machinery into a physical link between a displayed protein and the DNA encoding it, enabling large-scale selection ("panning"). George P. Smith, "Filamentous Fusion Phage: Novel Expression Vectors That Display Cloned Antigens on the Virion Surface" (Science 228, 1315-1317, 1985-06-14, OpenAlex W2070360032, 4,132 citations). Smith shared the 2018 Nobel Prize in Chemistry with Gregory Winter (who extended the technique to antibody libraries) and Frances Arnold. Directly enabled phage-display-based antibody discovery without hybridomas -- Humira itself was discovered this way -- and founded a standalone platform-licensing industry (Dyax, Cambridge Antibody Technology).
